Subject: Port location in relation to sub

Hi -
My plan is for a floor standing ported enclosure, roughly 12x12x48.
Before I finalize my plans I'd like to find out if there's a general rule for where to locate the sub in relation to the port.
It will probably be going in the corner of a pretty small room. I believe it's 15x11 or something in that range.
Should the sub downfire? Into a wall?
I've often heard the sub and port should be on the same face, but I don't see that applied much in home subs.
Can I have the sub toward the bottom of the enclosure and the port toward the top?

Or does any of this make any difference at all?
